1.运行软件,选择SYBR Green(R) I (如果使用taqman探针,则选择dual labeled probe程序)

2.根据管子选择平头还是凸头管

3.选择fam/sybr,edit profile,hold处选择时间和温度,如94度8分钟

4.cycling处设置变性,退火,延伸温度和时间,可以直接用鼠标拉动边缘调节(如果使用taqman探针则用两步法,94度15秒,60度1min).循环数45-50

5.点击start run,选择保存位置,开始运行

6.可以看运行所需时间,xx minutes remaining,时间到自动结束,打开保存文件分析即可
